Wolf G2 Cell Sorter Features

The WOLF was originally created by a team of scientists and engineers who wanted to solve a classic challenge in biological research: how to sort cells effectively, easily, and of high quality. Now, as science continues to move forward into increasingly complex realms, the WOLF is moving forward with it.

The new WOLF G2 instrument has significantly expanded the capabilities of gentle benchtop microfluidic cell sorting with two lasers and up to nine colors, while maintaining simple workflows for either bulk sorting or single-cell dispensing.

Single-cell sorting can be completed in 96- or 384-well plates when using the WOLF G2 in conjunction with the N1 Single-Cell Dispenser. This flexibility in performance, along with the additional abilities of the second laser, makes it ideal for use in many different research fields and application areas like single-cell genomics, cell line development, gene editing, antibody discovery, immunology, infectious disease, basic research, and more.

With two lasers and up to nine fluorescent channels, the WOLF G2 aligns with a broad set of research applications and experiments. Three different laser configurations allows options specific to your needs.

Catridge-based Fluidics

filler image

NanoCellect utilizes unique and proprietary microfluidic sorting cartridges for bulk sorting and single-cell dispensing. The cartridges employ a piezoelectric actuator and operates at less than 2 psi allows for gentle sorting that preserves viability and RNA integrity. Cartridges have a 100% disposable fluidic pathway, making it ideal for labs with many samples where cross-contamination or biohazard cleanup is an issue. The microfluidic cartridges are sterile and produce no aerosols, making it some of the safest sorting on the market. The cartridges also allow for maximum flexibility by allowing users to sort using their own buffers and media and only consume about 50 mL of sheath fluid in 1 day.

Product Specs Table Optics

The WOLF G2 uses laser-based flow cytometry technology for detecting and sorting a variety of cell types, including mammalian, plant, fungal, and bacterial cells. The WOLF G2 utilizes typical Forward- (FSC) and Back- (BSC) scatter detectors—as well as 3 different laser configurations to broaden its capabilities. In combination with the N1 single cell dispenser, scientists can complete both bulk and single-cell sorting. A simple cytometer, with powerful impact, allows researchers to sort their own cells on their own bench.

Available Laser Configurations:

  • 488 nm & 637 nm
  • 488 nm & 561 nm
  • 405 nm & 488 nm

Software

  • WOLFViewer software walks users though a simple workflow by offering step-by-step wizards to make the sorting process as straightforward as possible. As a result, the software uses an intuitive workflow allowing users to quickly complete setup.
  • After setup, WOLFViewer makes analyzing and creating gates simple—while still providing full-featured flow cytometry tools.
  • Key tools like automatic compensation and index sorting, are streamlined and included.
  • When sorting is complete, shutdown and cleanup is literally less than a minute, allowing users to rapidly conclude their work.
  • Researchers have the option of 2-way sorts for bulk sorting and 1-way sorts for single-cell dispensing through the WOLFViewer software.

Performance

The WOLF G2’s performance specifications allow users to complete various types of sorts for a variety of applications that may be occurring in their lab, for the ultimate in flexibility. The WOLF G2 can resolve lymphocytes, monocytes, and granulocytes, as well as microbial cells down to 1 μm, and up to 60 μm. Users can expect a sorting speed of up to 200 events per second maximally. Researchers have the option of 2-way sorts for bulk sorting and 1-way sorts for single-cell dispensing.

Antibody Discovery

The antibody drug discovery process differs from the traditional drug discovery of small molecules as it requires the development of hybridomas and the engineering of antibodies to recognize a specific target protein. The production of recombinant antibodies in cancer treatment as well as the development of neutralizing antibodies for infectious diseases, has allowed antibody discovery platforms to interrogate the immune repertoire and develop novel antibody libraries using flow cytometry analysis and cell sorting. The WOLF cell sorter provides a simple, gently cell sorting solution for many workflows such as:

  • Antibody Discovery from Single Bovine B-Cells
  • Isolate Antibody-Secreting CHO Cells
  • Recombinant protein production in CHO Cells

Cell Line Development

Generating cell lines has often been inefficient and affected by inaccurate limited dilution methods or the shear stress caused by conventional flow cytometer and traditional high-pressure cell sorters. NanoCellect Biomedical provides cell sorting solutions that allow to sort cells and establish stable cell lines using gentle low-pressure microfluidics. The WOLF’s ability to protect cell viability after sorting makes it an integral part of any cell line development workflow such as:

  • Recombinant protein production in CHO Cells
  • Sorting stem cell cultures
  • Antibody discovery

Gene Editing

Isolating edited-pluripotent stem cells with traditional sorting techniques can subject the cells to rough conditions in which gene expression, genomic stability and overall cell-health can become compromised. Microfluidic cell sorting with the WOLF, along with single cell deposition using the N1 single cell dispenser, eliminates the challenges that pluripotent stem cells face by sorting at gentle pressures without decompression shock and shear stress, and allow for efficient single cell deposition for high monoclonal outgrowth. Learn more about how cell sorting can improve workflows such as:

  • CRISPR and disease modelling
  • Protection of Karyotype
  • Generation of Genome-Edited iPSCs
